  • 細胞會感知外在環境的物理性訊號,改變細胞內分子活性以調整細胞骨架張力,甚至基因表現等來因應外在環境變化,此過程稱之為應力訊息傳遞。我實驗室的研究主要著重於細胞骨 架的調控,過去我們成功地建立了ROCK分子在細胞黏著點調控得分子機制,我們進而發現ROCK訊息的調控對於腎臟足細胞扮演重要角色。 足細胞是一種貼附在腎絲球基質的特化細胞,他們以特殊的足突結構交錯地包覆腎小球血管外圍,形成裂隙隔膜用來避免血液過濾時蛋白質的流 失;而足細胞的結構受損甚至脫落是導致慢性腎臟病惡化的重要關鍵。我們利用可條件誘發分化的小鼠足細胞株為模式,以 CRISPR/Cas9的方法建立基因剔除足細胞株,證實了ACTN4/Shp2/ROCK2的訊息傳遞對足細胞的附著力、細胞內張力與 過濾功能的重要性。我們建立了模擬足細胞進行過濾時的體外過濾裝置,以研究過濾液所造成的流體剪力刺激對足細胞的力訊息傳遞與細胞骨架變 化。此外,我們也與榮總腎臟科團隊合作,從基礎到臨床探討與腎絲球病變有關的基因與分子機制。
