  • 肝癌高居台灣癌症發生率第4名及死亡率第2名,其發生率逐年增加,但預後卻很差。儘管已有一些新的治療方法,但卻僅對一部分的患者有效,且常出現抗藥性,因此,急於開發更有效的療法。我們的主要研究是探討肝癌的致病機轉並提供有效的治療方式。藉由過去我們成功建立的CRISPR活體高通量篩選平台,來全面的找出參與肝癌形成的關鍵分子。另外,我們也會利用嶄新的肝癌類器官的培養方式,來驗證這些分子是否真的參與肝癌的生長,並且探討其在腫瘤進程中所扮演的角色以及其調控機制。此方法的優點是可以真實呈現腫瘤細胞在病人體內的情形,並可轉殖入老鼠體內,提供了一個藥物篩選平台,作為精準醫學研究之用,藉此可以找到更有效的藥物,以提供更好的治療效果。利用此二技術,我們可以更貼近生理情況下,尋找及證實這些分子的功能,更進一步地去測試這些分子的抑制劑是否可以達到治療的效果。
